Comments | To generate tissue for the developing anther and carpel (cLED) libraries we collected flowers from about 25 TA496 plants every couple of days for about a month. Flowers were approximately 5 days pre-anthesis (based on tagging the earliest stages we worked with - flower buds still expanding and sepals still tight together) through 5 days post anthesis - again based on tagging a few and collecting the rest based on similar appearance. At 5 dpa the carpels were about 0.5 cm in diameter. Flowers were collected at 5 different stages - 1) 5 days pre-anthesis, 2) anthesis (based on partial sepal seperation and anther tube swelling), 3) 5 dpa, 4) intermediate between 1 and 2, and intermediate between 2 and 3. It was obtained about 0.5 g of the earlier stages from hundreds of flowers while 10 or so fruit of the #5 stage gave us a gram. I was combined all of this (0.5 - 1 g per stage) andused that for RNA isolation. These carpels included ovules and embryos.The anthers from the first 3 stages and some of the 4th (that still had bthem) were collected for the anther library. |
