Comments | The EcoRI site was destroyed during cloning. Use any enzyme to the left of it along with XhoI to cut out the insert. Also note that the library was amplified prior to mass excision 11,520 clones were picked into 30 x 384-well plates. The primary library titre was 106 pfu/ml and the amplified library titre is 109 pfu/ml. About 108pfu/ml were excised. The sample is a mix of leaves treated with 2,6 dichloroisonicotinic acid (0.5mM INA by foliar spray aplication or petiole immersion-treat in solution), benzothiadizole (1.5 mM BTH by foliar spray aplication or petiole immersion-treat in solution), jasmonic Acid (100 uM JA by petiole immersion in closed chamber incubation), ethylene induced xylanase from fungus Trichoderma (1 ug/ml of EIX by vacuum infiltration), Fenthion (0.001% by vacuum infiltration), Systemin (1 nM by petiole immersion) and Okaidaic Acid (1uM by transpiration stream for 45 min) |
